The Ocean Stewardship Fund is a program held by the marine stewardship council (MSC) in an effort to help promote sustainable fisheries. This program is specifically for academics or an organization engaged in fisheries management around the world. Anggini Fuji Astuti with research related to Stock Status and Distribution of Secondary Species Using Environmental Biological and DNA Assessment in the Crayfish (Portunus sanguinolentus) Fishery on Madura Island received funding from OSF. This data collection is also important for the Indonesian Blue Swimming Crab (APRI) regarding the fulfilment of indicators to minimize environmental impacts around the crab fishery.
Environmental DNA (eDNA) is a new approach to assessing marine biodiversity that can address the challenges of conventional monitoring and complement existing biodiversity assessment surveys. As technology develops, the technique of using DNA metabarcoding has become an important aspect of biodiversity research. Environmental DNA metabarcoding can provide a broader assessment of biodiversity than conventional visual census methods. Environmental DNA samples taken in this study were water samples at 13 station points surrounding the island of Madura which will be analysed at the Oceanogen Laboratory. Considering the potential of the Portunus sanguinolentus species, it is necessary to research on the distribution stock status and stock units of the Portunus sanguinolentus species on Madura Island. In addition to meeting the need for information regarding stock assessment and effective management of crab fisheries, it is necessary to conduct research on its biological aspects.

Information on the stock structure of the Portunus sanguinolentus species is useful for determining the right conservation unit to estimate stock composition in the fishery. In fulfilling this information, the Truss Network Analysis method is used, which is a fishery management analysis system that is able to verify a large number of samples in a short time. Truss Network Analysis is also used in the same series of measurements to investigate the separation of stock units within possible species. In addition, conventional measurements were carried out measuring the width of the shell (CW) and weight to determine the status of the crab stock status of Portunus sanguinolentus in Madura Island.

This research works in collaboration with international organizations such as MSC who can help add expertise around the data needed for sustainable fisheries, with OSF helping to facilitate DNA research. Then this research collaborated with the Indonesian Crab Management Association (APRI). Starting from the beginning of September, research on Stock Status and Distribution of Secondary Species Using Environmental Biological and DNA Assessment in Crayfish (Portunus sanguinolentus) Fisheries on Madura Island has been carried out and is still ongoing until now. The first research location visited on Madura Island was Bangkalan Regency, where this area has a community with the majority of the population working as fishermen.
The type of crab, Portunus sanguinolentus, is not the target of catching fishermen in Bangkalan Regency. This is because the comparison of the price of the jaul crab Portunus sanguinolentus is much cheaper than the crab jaul Portunus pelagicus. The fishing process carried out by fishermen does not stay in one location. Usually they will make arrests at several different points. This is because to avoid the catch in the form of crabs of the type Portunus sanguinolentus which has low economic value. This is one of the obstacles in the process of collecting data for the Portunus sanguinolentus crab so that the data obtained is limited. (Anggini/IPB)
